What is Sofosbuvir & Ledipasvir Tablets?

  • Sofosbuvir:
    • Sofosbuvir is an antiviral medication that belongs to a class of drugs known as nucleotide polymerase inhibitors.
    • It works by interfering with the replication process of the hepatitis C virus, inhibiting its ability to multiply in the liver.
    • Sofosbuvir was a significant advancement in HCV treatment, as it was one of the first direct-acting antiviral drugs approved for use.
  • Ledipasvir:
    • Ledipasvir is also an antiviral medication and belongs to a class of drugs known as HCV NS5A inhibitors.

  • Treatment of Hepatitis C Genotypes:
    • Sofosbuvir and Ledipasvir combination therapy is particularly effective against specific genotypes of the hepatitis C virus. The most common genotype treated with this combination is genotype 1, but it can also be used for other genotypes in certain cases.
  • High Cure Rates:
    • This combination therapy has shown high cure rates, often defined as achieving sustained virologic response (SVR), which means the virus is no longer detectable in the blood after completing the treatment course.
  • Minimizing Liver Damage:

Sofosbuvir & Ledipasvir Tablets – Mechanism of Action and Pharmacology

Mechanism of Action:

  • Sofosbuvir:
    • Sofosbuvir is a nucleotide analog inhibitor of the HCV NS5B polymerase.
    • It works by being converted into its active form (triphosphate) inside the liver cells. This active form then competes with the natural building blocks (nucleotides) required for the replication of the HCV RNA.
    • When the active form of sofosbuvir is incorporated into the growing HCV RNA chain, it causes premature termination of the chain, effectively inhibiting the ability of the virus to replicate and produce new copies of itself.
  • Ledipasvir:
    • Ledipasvir is an inhibitor of the HCV NS5A protein, which plays a crucial role in the replication process of the hepatitis C virus.
    • NS5A is involved in several steps of the viral lifecycle, including RNA replication and virion assembly.
    • By inhibiting the function of NS5A, ledipasvir disrupts multiple stages of the viral replication cycle, ultimately suppressing the production of new virus particles.
